First tangible evidence of e=mc2 was not at nuclear testing. It was evident in the early days of radioactive testing from Curie to Roentgen and Bohr.

e=mc2, you did not even explain what this meant.
Energy = Mass x Speed of light squared.
The speed of light being 3x10^8
This means that when a critical reaction of U-238 occurs, and it decays to U-235. This means it loses 3 neutrons per atom. This small loss adds up, 3 grams every 238grams of uranium in a warhead. Where does it go?
To energy.
